- Immunofluorescent analysis of C1orf77 (red) and Insulin (green) were performed on mouse pancreatic tissue from NOD (Non-obese diabetic) mice and CD1 (cluster of differentiation 1) mice. To expose target proteins, antigen retrieval was performed by microwaving tissues for three times with each 5 minutes in 10mM sodium citrate buffer (pH 6.0). Following antigen retrieval slides were wash with PBS and PBS+1% lamb serum, and then the slides were blocked with 1:50 normal donkey serum for 30 min at room temperature. The tissues were probed for C1orf77 (Product # PA5-44307) antibody 1:200 dilution and Insulin overnight at 4°C in a humidified chamber. Tissues were washed with PBS and PBS+1% lamb serum for 5 min and detection was performed using an fluorescence-conjugated secondary antibodies. Images were obtained using Zeiss confocal microscopy at 40X magnification.
