- FOXO3A Monoclonal Antibody (K.115.9)
- Antibody type
- Monoclonal
- Antigen
- Synthetic peptide
- Description
- Antibodies to this protein (and modification) were previously sold as part of a Thermo Scientific Cellomics High Content Screening Kit. This replacement antibody is now recommended for researchers who need an antibody for high content cell based assays. It has been thoroughly tested and validated for cellular immunofluorescence (IF) applications. Further optimization including the selection of the most appropriate fluorescent Dylight conjugated secondary antibody may have to be performed for your high content assay.

- Knockout of FOXO3 was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID CRISPR979902_LV) and LentiArray Cas9 Lentivirus (Product # A32064). Western blot analysis of FOXO3 was performed by loading 30 µg of HeLa wild type (Lane-1), HeLa Cas9 (Lane-2) and HeLa FOXO3 KO (Lane-3). The samples were electrophoresed using NuPAGE™ Novex™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with FOXO3A Monoclonal Antibody (K.115.9) (Product # MA5-14932, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10,000 dilution) using the iBright™ FL1500 (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). Loss of signal upon CRISPR mediated knockout (KO) using the LentiArray™ CRISPR product line confirms that antibody is specific to FOXO3.

- Figure 3 Primordial follicle activation with PPARgamma modulation. ( A ) Nuclear exclusion of forkhead box O3a (FOXO3a) (red) in oocytes of primordial follicles 6 h after treatment with PPARgamma modulators. Arrow heads point to FOXO3a located in the nuclei, whereas arrows point to FOXO3a located in the cytoplasm. Anti-Mullerian hormone (AMH) and Ki-67 staining are also shown in red, as indicated. Counterstaining was performed using anti-actin antibodies (green) and nucleus with DAPI (blue). Scale bar = 10 mum. ( B ) Percentages of primordial follicles with nuclear export of FOXO3a. ( C ) In vitro culture for 12 days of 5-day-old ovaries after 48 h treatment of PPARgamma modulators. Scale bar = 100 mum. ( D ) Western blot analysis of ovaries after a 3-day treatment with PPARgamma modulators. ( E ) Quantitation of results shown in ( D ). * indicates significant differences between groups, p < 0.05.
