Experimental details: Western blot analysis was performed on whole cell extracts (30 µg lysate) of A549 (Lane 1), DU-145 (Lane 2), PC-3 (Lane 3), LNCaP (Lane 4) and Caco-2 (Lane 5). The blot was probed with Anti-CYP27A1 Polyclonal Antibody (Product # PA5-27946, 1:1000 dilution) and detected by chemiluminescence using Goat anti Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 72 kDa band corresponding to CYP27A1 was detected across the cell lines tested and the protein expression was found to be low in LNCaP when compared to PC-3.

Experimental details: Knockdown of CYP27A1 was achieved by transfecting A549 cells with CYP27A1 specific siRNAs (Silencer® select Product # s3887, s3888). Western blot analysis (Fig. a) was performed using whole cell extracts from the CYP27A1 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with CYP27A1 Polyclonal Antibody (Product # PA5-27946, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to CYP27A1.
