- Description: The N28.1 monoclonal antibody recognizes the Fc receptor homolog expressed in B cells (FREB), also know as FCRLA or FcRX. FREB is a member of the family of Fc receptors for IgG (FcRg). Unlike FcRg, FREB lacks a transmembrane sequence and has an intracellular localization. FREB has been detected in proliferating and activated B cells, EBV-transformed B cells, germinal center centroblast, follicular lymphoma and the Burkitt's lymphoma cell line Raji. In contrast with published reports, the N28.1 antibody was also able to detect expression of FREB in resting B cells from normal human blood samples. Applications Reported: This N28.1 antibody has been reported for use in intracellular staining followed by flow cytometric analysis, and immunohistochemical staining. (Fluorochrome conjugated N28.1 is recommended for use in intracellular flow cytometry). Applications Tested: This N28.1 antibody has been tested by intracellular staining followed by flow cytometric analysis of normal human peripheral blood cells. This can be used at less than or equal to 0.03 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. IC Fixation Buffer (Product # 00-8222-49) and Foxp3/Transcription Factor Staining Buffer (Product # 00-5523-00) have been tested for use with the N28.1 antibody and gave identical results. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2 µm post-manufacturing filtered.
