- Western blot analysis of Phospho-beta-Catenin pSer552 in total cell lysates from SK-N-MC cells, treated with forskolin (FSK) for 30 minutes, or lysate treated with lambda phosphatase for 1 hour, using Phospho-beta-Catenin pSer552 polyclonal antibody (Product # PA5-17154) (upper) or a Beta-Catenin polyclonal antibody (bottom).

- Western blot analysis was performed on whole cell extract (30 µg lysate) of A-431 (Lane 1), A-431 treated with EGF (100 ng/mL, 6hr) (Lane 2). The blot was probed with Anti-Phospho-beta Catenin (Ser552) Polyclonal (Product # PA5-17154, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 90 kDa band corresponding to Phospho-beta Catenin (Ser552) was observed in A-431 and was enhanced upon cell treatment.

- Western blot analysis of phospho beta catenin was performed by loading 30 µg of HT29 WT untreated (Lane 1), HT29 WT EGF treated (Lane 2), HT29 Cas9 untreated (Lane 3), HT29 Cas9 EGF treated (Lane 4), HT29 beta catenin knockout untreated (Lane 5) and HT29 beta catenin knockout EGF treated (Lane 6) whole cell lysate. The blot was probed with Phospho-beta Catenin (Ser552) Polyclonal Antibody (Product # MA5-17154, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution. Loss of signal upon CRISPR mediated knockout (KO) confirms that antibody is specific to phospho beta catenin. A non-specific band was also observed at ~52 kDa with the above testing conditions.
