- Western blot analysis of MR1 in multiple sample types using a MR1 polyclonal antibody (Product # PA5-98768) at a dilution of 1:2,000. A goat anti rabbit polyclonal secondary antibody was used at a dilution of 1:50,000. Sample types that were tested include: THP-1 whole cell lysate, K-562 whole cell lysate, Raji whole cell lysate, Hela whole cell lysate, A2780 whole cell lysate, and mouse brain tissue.

- Western blot was performed using Anti-MR1 Polyclonal Antibody (Product # PA5-98768) and 32-55 kDa bands corresponding to Major histocompatibility complex class I-related gene protein were observed across cell lines tested. Whole cell extracts (30 µg lysate) of Jurkat (Lane 1), Raji (Lane 2), HeLa (Lane 3), Daudi (Lane 4) and Caco-2 (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Reagent Kit (Product # WP20005).

- Immunofluorescence analysis of Major histocompatibility complex class I-related gene protein was performed using HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with MR1 Polyclonal Antibody (Product # PA5-98768) at 1:100 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790), (1:2000 dilution), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b:Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300 dilution). Panel d represents the merged image showing cytoplasmic (ER, endosome, golgi) and membrane localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

- Immunohistochemistry (Paraffin) analysis of MR1 in paraffin-embedded human liver tissue. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 min at RT, then the primary antibody (1% BSA) was incubated at 4°C overnight. The sample was incubated with a MR1 polyclonal antibody (Product # PA5-98768) at a dilution of 1:200, followed by a goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
