- OMA1-06020 detects smoothelin from human, canine, feline, porcine, chicken and monkey tissues. This antibody detects both the visceral and vascular smoothelin isoforms. Does not cross react with rat samples. OMA1-06020 has been successfully used in Western blot, immunocytochemistry, and immunohistochemistry procedures. By Western blot, this antibody detects an ~59 kDa protein representing visceral smoothelin and an ~100 kDa protein representing vascular smoothelin from canine smooth muscle extracts. This antibody can be used to discriminate fully differentiated (contractile) smooth muscle cells from proliferative smooth muscle cells. Staining of formalin-fixed paraffin-embedded tissues requires boiling in 10 mM citrate buffer (pH 6.0) for 3x5 min, prior to immunostaining. OMA1-06020 antigen is cytoskeletal smoothelin extract of chicken gizzard. Store at 4ºC, or in small aliquots at -20ºC.

- Western blot was performed using Anti-Smoothelin Monoclonal Antibody (R4A) (Product # OMA1-06020) and a 100kDa band corresponding to Smoothelin was observed across cell lines and tissues tested . Nuclear enriched extracts (30 µg lysate) of PC-3 (Lane 1), HeLa (Lane 2), Mouse Uterus (Lane 3), Mouse Brain (Lane 4) and Mouse Lung (Lane 5) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:5000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
