- Description: The monoclonal antibody CF-145 recognizes human S100A8. S100A8, also known as migration inhibitory factor-related protein 8 (MRP-8) or Calgranulin-A, is a calcium binding protein and member of the S100 family of proteins. S100A8 is expressed by neutrophils, monocytes, macrophages, granulocytes, and epithelial cells during dermatoses, such as psoriasis. S100A8 forms a heterodimer with S100A9, which together is known as calprotectin. S100A8 has antimicrobial and pro-apoptotic properties and is a pro-inflammatory mediator in acute and chronic inflammation. Levels of S100A8 are elevated in patients with various inflammatory disorders such as cystic fibrosis, rheumatoid arthritis, and chronic bronchitis. Upregulation of S100A8 is also observed in many tumors, including skin, breast, colon, pancreatic, and ovarian cancers. Applications Reported: This CF-145 antibody has been reported for use in flow cytometric analysis, and intracellular staining followed by flow cytometric analysis. Applications Tested: This CF-145 antibody has been pre-titrated and tested by intracellular staining followed by flow cytometric analysis of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Please refer to Best Protocols: Protocol A: Two step protocol for (cytoplasmic) intracellular proteins located under the Resources Tab online. This can be used at 5 µL (0.06 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. eFluor® 660 is a replacement for Alexa Fluor® 647. eFluor® 660 emits at 659 nm and is excited with the red laser (633 nm). Please make sure that your instrument is capable of detecting this fluorochome. Excitation: 633-647 nm; Emission: 668 nm; Laser: Red Laser. Filtration: 0.2 µm post-manufacturing filtered.

- Normal human peripheral blood cells were incubated with Human Fc Receptor Binding Inhibitor Purified (Product # 14-9161-73) for 20 minutes, then stained with Anti-Human CD11b PE (Product # 12-0118-42) followed by fixation and permeabilization with the Intracellular Fixation and Permeabilization Buffer Set (Product # 88-8824-00) and protocol. The cells were then stained with Mouse IgG2b K Isotype Control eFluor® 660 (Product # 50-4732-82) (left) or Anti-Human S100A8 eFluor® 660 (right). Cells in the monocyte gate were used for analysis.
