Detection of human CEP170 by western blot. Samples: Whole cell lysate (15 µg) from HEK293T, A-549, K-562, and HeLa cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-CEP170 antibody (Product # A301-024A; lot A301-024A-2) used for WB at 0.04 µg/mL. Detection: Chemiluminescence with an exposure time of 3 minutes.

Experimental details

Detection of human CEP170 by western blot and immunoprecipitation. Samples: Whole cell lysate (5, 15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-CEP170 antibody A301-024A used for WB at 0.04 µg/ml (A) and 1 µg/ml (B) and used for IP at 3 µg/mg lysate. CEP170 was also immunoprecipitated by rabbit anti-CEP170 antibodies A301-023A and BL4463, which recognize upstream epitopes. Detection: Chemiluminescence with exposure times of 30 seconds (A and B).

No. A301-024A) used at a dilution of 1:200 ( 1 µg/mL). Detection: Red-fluorescent goat anti-rabbit IgG highly cross-adsorbed Antibody used at a dilution of 1:100.

Detection of human CEP170 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-CEP170 antibody (Product # A301-024A; lot A301-024A-2) used for IP at 6 µg per reaction. CEP170 was also immunoprecipitated by a previous lot of this antibody (lot A301-024A-1) and rabbit anti-CEP170 antibody BL4462. For blotting immunoprecipitated CEP170 (Product # A301-024A) was used at 0.04 µg/mL. Detection: Chemiluminescence with an exposure time of 10 seconds.
