Description

PA1-1752 detects presenilin 1 protein (PS1) from human, rat and mouse samples. PA1-1752 has successfully been used in Western blot procedures. By Western blot, this antibody detects an ~66 kDa protein representing PS1. The PA1-1752 immunogen is a synthetic peptides corresponding to residues K(311) N S K Y N A E S T E R(322) and E(342) A Q R D S H L G P H R(353) of human PS1.

Experimental details

Western blot analysis was performed on membrane enriched extracts (30 µg lysate) of MKN45 (Lane 1). The blot was probed with Anti-Presinillin 1 Chicken Monoclonal Antibody (Product # PA1-1752, 1 µg/mL) and detected by chemiluminescence using Goat anti-Chicken IgY (H+L) Secondary Antibody, HRP conjugate (Product # A16054, 1:4000 dilution). Two isoforms of 53 and 42 kDa corresponding to Presinillin 1 was observed across the cell line tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE&reg ; 10% Bis-Tris gel (Product # NP0302BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
