Description: Description: The 2H7 monoclonal antibody reacts with human CD20, a 33-36 kDa transmembrane protein. CD20 is expressed by developing B cells as well as mature B cells but not plasma cells. CD20 has been detected at low levels on a small subset of mature T cells. It is suggested that CD20 plays a role in B-cell activation. Applications Reported: This 2H7 antibody has been reported for use in flow cytometric analysis. Applications Tested: This 2H7 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. This may be used at 5 µL (0.06 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. Alexa Fluor 532 is excited with the Green laser (532 nm) and emits at 561 nm. This cannot be used with the Yellow-Green laser (561 nm). We recommend using a 560/14 band pass filter. Please make sure that your instrument is capable of detecting this fluorochrome. Excitation: 532 nm; Emission: 561 nm; Laser: Green Laser

Experimental details: Figure 6 Flow cytometry detection of TV antigens-containing cells in vitro . A- PBMCs isolated from healthy rhesus macaques were used for in vitro inoculation with TV. After being cultured in vitro for 6 days, cells were sorted out into CD3+ T cell and CD20+ B cell populations. Presence of TV antigens was detected in CD20+ B cells as shown by histograms (black peak). B- The CD20+ B cells were further subdivided into populations expressing the HLA-DR, CD11c or CD123 antigens. Presence of TV was revealed predominantly in CD20+HLA-DR+ cells while some of the other lymphocyte populations including the CD20+CD11c+ and CD20+CD123+ B cells also contained TV antigens.

Experimental details: Figure 8 T cell electroporation with Chimeric Antigen Receptor (CAR) results in stable gene expression conferring target-specific cytotoxicity. PBMCs from one donor were electroporated using 1SM buffer, 20 ug of pT3-20z plasmid and 0,5 ug of SB100x transposase plasmid. One day later, lymphocytes were stimulated with irradiated L388 cells and CAR expression was evaluated until d+30 by flow cytometry analysis using an anti-fab antibody.
