- Western blot analysis of c-Myc in whole cell protein from PC3 (lane 1), U-2 OS (lane 2) and mouse testis (lane 3). Samples were incubated in c-Myc monoclonal antibody (Product # MA1-16637) using a dilution of 2.0 µg/mL followed by an anti-mouse HRP secondary antibody. Separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. Detection: chemiluminescence.

- Western blot was performed using Anti-c-Myc Monoclonal Antibody (9E11) (Product # MA1-16637) by loading whole cell extracts of untransfected and transiently transfected HEK-293E lysates: untransfected, 60 µg (Lane 1), Myc-p65, 60 µg (Lane 2), Myc-p65, 30 µg (Lane 3), Myc-p65, 15 µg (Lane 4), Myc-p65, 7.5 µg (Lane 5),and 25 ng of Positope (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). A ~67 kDa band corresponding to Myc-p65-V5 observed in HEK293E transfected lysates on probing with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP (Product #A28177, 1:20,000 dilution). Positope (Product # R900-50) is a 53 kDa recombinant protein consisting of multiple epitope tags, which has been used as a positive control for c-Myc tag detection (Lane 6).

- Immunohistochemical analysis of c-Myc in immersion fixed paraffin-embedded sections of human breast cancer. Samples were incubated in c-Myc monoclonal antibody (Product # MA1-16637) using a dilution of 1:600 overnight at 4 °C. Tissue was stained using the VisuCyte anti-mouse HRP polymer detection reagent with DAB chromogen (brown) and counterstained with hematoxylin (blue).

- Flow cytometry of c-Myc in U937 cells. Samples were incubated in c-Myc monoclonal antibody (Product # MA1-16637) using a dilution of 2.5 µg/mL for 30 minutes at room temperature. Antibody (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Both antibodies were conjugated to Alexa Fluor 647.
