Description

Bifikafusp alfa is an investigational immunocytokine, not a traditional monoclonal antibody. It is a fusion protein consisting of the human L19 antibody fragment (scFv) genetically fused to the cytokine human interleukin-2 (IL-2). The L19 fragment specifically targets the extra domain B (EDB) of fibronectin, a protein marker of tumor angiogenesis that is abundantly expressed in the neovasculature of most solid tumors but is virtually absent in normal adult tissues. The IL-2 component is a potent T-cell growth factor. This design allows for the targeted delivery of IL-2 directly to the tumor microenvironment, aiming to locally activate and expand tumor-infiltrating lymphocytes (TILs) and natural killer (NK) cells while minimizing the severe systemic toxicity associated with high-dose recombinant IL-2 therapy. It has demonstrated anticancer activity in clinical trials and is being evaluated for the treatment of various advanced solid tumors.
