MA5-14812 detects Phospho-GAB1 pTyr627 in human samples. Predicted to react with mouse and rat based on sequence homology. MA5-14812 has been successfully used in Western blot applications. The MA5-14812 immunogen is synthetic phosphopeptide corresponding to residues surrounding Tyr627 of human Gab1. MA5-14812 detects Phospho-Gab1 (Tyr627) which has a predicted molecular weight of approximately 110 kDa.

Western blot was performed using Anti-Phospho-GAB1 (Tyr627) Monoclonal Antibody (Product # MA5-14812) and a 110 kDa band corresponding to Phospho-GAB1 (Tyr627) was observed to be enhanced upon treatment with EGF, Insulin and NGF across all the cell lines tested. Whole cell extracts (30 µg lysate) of 3T3-L1 (Lane 1), 3T3-L1 treated with EGF (200 ng/mL for 10 mins) (Lane 2), 3T3-L1 treated with Insulin (100 nm for 10 mins) (Lane 3), PC-12 (Lane 4), PC-12 treated with EGF (200 ng/mL for 10 mins) (Lane 5) and PC-12 treated with NGF (100 ng/mL for 10 mins) (Lane 6) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
