- Western blot analysis was performed on whole cell extracts (30 µg lysate) of A549 (Lane 1), COS-7 (Lane 2), MDCK (Lane 3), C2C12 (Lane 4), MDA-MB-231 (Lane 5), PC-12 (Lane 6), RSC96 (Lane 7) and tissue extracts of Mouse Lung (Lane 8). The blot was probed with beta Actin Polyclonal Antibody (Product # PA5-16914 1 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 42 kDa band corresponding to beta Actin was observed across the cell lines and tissues tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 10% Bis-Tris gel (Product # NP0322BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).

- Immunofluorescence analysis of beta Actin was performed using 70% confluent log phase MDA-MB-231 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with beta Actin Rabbit polyclonal antibody (Product # PA5-16914) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytoskeletal localization. Panel e shows the no primary antibody control. The images were captured at 60X magnification.

- Figure 5. NEUROD2 and STIM1 expression are inversely correlated across cortical development. A , B , Immunoblotting analysis and quantification of protein levels across development in cerebral cortical tissue revealed an inverse correlation between NEUROD2 and STIM1 protein expression. STIM1 protein levels are normalized to the amount of beta-actin. Data represents two biological replicates, each quantified as duplicates. Bars represent the SEM. C , Stim1 and Neurod2 mRNA levels were plotted as a function of age in human prefrontal cortex using postmortem tissue. Plots were acquired from braincloud.jhmi.edu ( Colantuoni et al., 2011 ). Similar to mouse data, an inverse correlation in Neurod2 and Stim1 expression was observed in humans as well.

- Figure 3 Semiquantitative immunoblotting of MBP and CPT1A. Protein expression was investigated in the HB of WT EAE (n = 6) and Cpt1a P479L EAE mice (n = 6). Immunoblotting with MBP revealed four bands of 14.0, 17.0, 18.5 and 21.5 kDa ( a ). All detected MBP isoforms from Cpt1a P479L EAE mice were expressed at significantly higher levels than those from WT EAE mice (two-way ANOVA with Sidak's multiple comparisons post hoc test) ( b ). Analysis of CPT1A protein ex p ression revealed no significant differences between WT EAE and Cpt1a P479L EAE mice ( c , d ). The data were normalized to the density of the beta-actin control. Data are presented as the mean +- SEM. Asterisks indicate the level of significance (**p < 0.01, ***p < 0.001, ****p < 0.0001). More information about full-length blots can be found in Supplementary Figs S1 and S2 .

- Figure 2 Effect of beta-NF and EtOH treatment on cytochrome P450 (CYP) isoforms' expression in undifferentiated SH-SY5Y cells. ( a - d ) The graphs show the relative protein levels quantification by Western blot for the CYP2D6 ( a ), 2E1 ( b ), 1A1 ( c ), and 3A4 ( d ) isoforms after treating cells with the inducers for 48 h (see Section 4 ). Data was acquired by measuring the fluorescent intensity per pixel on each band. The relative amount of protein normalised with beta-actin as a housekeeping protein for each condition was plotted as fold-increase and compared to the control, which was given a value of 1. Columns represent the mean +- SEM of at least three different experiments. Statistical significance was analyzed by one-way ANOVA followed by a Tukey post-test (* p < 0.05; ** p < 0.01). ( e ) Top panel shows a representative blot of beta-actin housekeeping protein detected with secondary antibody Cy3 (green) in control (2nd lane), beta-NF (3rd lane) and EtOH (4th lane) treatments. ( e ) Bottom panel shows representative blots of each isoform detected with secondary antibody Cy5 (red) in the mentioned conditions.
