- Western blot was performed using Anti-TAGLN Monoclonal Antibody (GT4511) (Product # MA5-17288) and 23kDa band corresponding to TAGLN was observed across all cell lines and tissues tested except Jurkat, Raji, Rat Skeletal Muscle and Rat Brain which are known to be low expressing (DOI: 10.1006/geno.1998.5267, 10.1371/journal.pone.0156429). Whole cell extracts (50 µg lysate) of BJ (Lane 1), Caco-2 (Lane 2), HeLa (Lane 3), Jurkat (Lane 4), Raji (Lane 5), Rat Urinary Bladder (Lane 6), Rat Uterus (Lane 7), Rat Skeletal Muscle (Lane 8) and Rat Brain (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). Expression of TAGLN was observed in BJ, Caco-2, HeLa, Rat Urinary Bladder and Rat Uterus but not in Jurkat, Raji, Rat Skeletal Muscle and Rat Brain which are known to be low expressing.
