Urelumab is a fully human monoclonal antibody agonist targeting CD137 (4-1BB), a co-stimulatory receptor on activated T cells and NK cells. It binds to CD137 to enhance T cell proliferation, cytokine production, and cytotoxic activity, thereby boosting anti-tumor immunity. It was investigated for the treatment of advanced solid tumors and hematologic malignancies, but clinical development was limited due to dose-dependent hepatotoxicity.
