- Western blot analysis of PCNA was performed by loading 50 µg of the indicated whole cell lysates per well, and 10 µL of PageRuler Plus Prestained Protein Ladder (Product # 26619) onto a 4-20% Tris-HCl polyacrylamide gel. Proteins were transferred to a PVDF membrane using the G2 Fast Blotter (Product # 62288), and blocked with StartingBlock T20 (TBS) Blocking Buffer (Product # 37543) for 1 hour at room temperature. PCNA was detected between 30-35 kDa after probing with a PCNA monoclonal antibody (Product # MA5-11358) at a dilution of 1:50 in StartingBlock T20 (TBS) Blocking Buffer (Product # 37543) overnight at 4C on a rocking platform, followed by an HRP-conjugated goat anti-mouse IgG secondary antibody (Product # 31430) at a dilution of 1:20,000 for 1 hour. Chemiluminescent detection was performed using SuperSignal West Dura (Product # 34075). Images were acquired on a Thermo Scientific myECL Imager (Product # 62236).

- Knockdown of PCNA was achieved by transfecting HeLa with PCNA specific siRNAs (Silencer® select Product # S10133, S10134). Western blot analysis (Fig. a) was performed using Nuclear enriched extracts from the PCNA knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with PCNA Monoclonal Antibody (PC10) (Product # MA5-11358, 1:50) and Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to PCNA.

- Western blot was performed using Anti-PCNA Monoclonal Antibody (PC10)(Product # MA5-11358) and a 32kDa band corresponding to PCNA was observed across all tested cell lines. Nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), MCF7 (Lane 2), Jurkat (Lane 3), HEK-293 (Lane 4), NIH/3T3 (Lane 5), MDA-MB-231 (Lane 6), A549 (Lane 7), SH-SY5Y (Lane 8), PC-12 (Lane 9) were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel (Product # NP0342BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody ( 1:50) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunofluorescent analysis of PCNA (purple) in HeLa cells. The cells were fixed and permeabilized with ice-cold methanol for 15 minutes, and blocked with 1% Blocker BSA in PBS (Product # 37525) for 15 minutes at room temperature. Cells were stained with a PCNA monoclonal antibody (Product # MA5-11358) at a dilution of 1:50 in 1% Blocker BSA in PBS (right panel) or incubated in blocking buffer alone as a negative control (left panel) for 1 hour at room temperature, and then incubated with a DyLight 650-conjugated goat anti-mouse IgG secondary antibody (Product # 84545) at a dilution of 1:250 for 30 minutes at room temperature. Actin (green) was stained with a DyLight 488-conjugated beta-actin monoclonal antibody (Product # MA5-15739-D488) and nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ToxInsight Instrument at 20X magnification.

Cells were harvested, adjusted to a concentration of 1-5x10^6 cells/mL, fixed with 2% paraformaldehyde and washed with PBS. Cells were blocked with a 2% solution of BSA-PBS for 30 min at room temperature and incubated with a PCNA monoclonal antibody (Product # MA5-11358) at a dilution of 1:2 for 40 min at room temperature. Cells were then incubated for 40 min at room temperature in the dark using a Dylight 488-conjugated secondary antibody and re-suspended in PBS for FACS analysis.

- Figure 3 Self-renewal of neural progenitors in cerebellum was not abrogated upon Sox9 inactivation as revealed by immunofluorescence staining on cerebellar sagittal sections. (A-D) Expression of the neural stem cell marker Sox2 was not lost in the Sox9- cerebellum during late gestation under both En1-Cre and Pax2-Cre mediated Sox9 inactivation. In both groups, scattered signals were observed in the VZ and the prospective white matter, while the EGL displayed the strongest signal which corresponded to the actively proliferating granule cell precursors. (A') and (B') showed enlarged view of the VZ. (E-J) Quantification of mitotic activity at the VZ by double pulses of BrdU at either E16.5 (E-F) or E18.5 (G-H) did not reveal any difference in the proliferation of VZ neural progenitors between control and the Pax2-Cre induced Sox9 - mutant.
