- Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), PC-3 (Lane 2), F9 (Lane 3), RAW 264.7 (Lane 4), K-562 (Lane 5) and Neuro-2a (Lane 6). The blot was probed with Anti-RANGAP1 Polyclonal Antibody (Product # PA1-31482, 1:3000 dilution) and detected by chemiluminescence using Rabbit anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27014, 0.25 µg/ml, 1:4000 dilution). A 63 kDa band corresponding to RANGAP1 was observed across the cell lines tested.

- Western Blot analysis of RANGAP1 was performed by loading 35 µg (in RIPA buffer) of 3T3 lysates. Proteins were transferred to a membrane and probed with a RANGAP1 Polyclonal Antibody (Product # PA1-31482) at a dilution of 0.2 µg/mL.

- Knockdown of RANGAP1 was achieved by transfecting PC-3 cells with RANGAP1 specific siRNAs (Silencer® select Product # s11776, s11777). Western blot analysis (Fig. a) was performed using whole cell extracts from the RANGAP1 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with RANGAP1 Polyclonal Antibody (Product # PA1-31482, 0.5µg/ml) and Rabbit anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27014, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to RANGAP1.
