- Immunoprecipitation was performed on HEK293T cells overexpressing GAD1. Antigen-antibody complexes were formed by incubating the sample with 2 µg of GAD1 monoclonal antibody (Product # MA5-24909) antibody and 0.1 mg of goat anti-mouse conjugated magnetic beads overnight. The sample was washed extensively and probed with rabbit anti-DDK polyclonal antibody.

- Western blot was performed using Anti-GAD67 Monoclonal Antibody (OTI3G9) (Product # MA5-24909) and a 67kDa band corresponding to GAD67 was observed in Mouse Brain and Rat Brain. No expression was observed in Mouse Ovary, Rat Ovary and Mouse Kidney which are known to be negative for GAD67. Tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Rat Brain (Lane 2), Mouse Ovary (Lane 3), Rat Ovary (Lane 4) and Mouse Kidney (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).An uncharacterized band (*) was observed at ~58kDa. Band corresponding to Mouse IgG light chain was observed at ~25kDa.
