Experimental details

SIN3A Polyclonal Antibody detects SIN3A protein at nucleus by immunofluorescent analysis. Sample: HeLa cells were fixed in 4% paraformaldehyde at RT for 15 min. Green: SIN3A stained by SIN3A Polyclonal Antibody (Product # PA5-85554) diluted at 1:1,000. Red: phalloidin, a cytoskeleton marker, diluted at 1:200. Scale bar= 10 µm.

Cross-linked ChIP was performed with MCF-7 chromatin extract treated with B-estradiol (10 nM for 45 min) and 5 µg of either control rabbit IgG or SIN3A Polyclonal Antibody (Product # PA5-85554). The precipitated DNA was detected by PCR with primer set targeting to GAPDH or MyoD.

Immunoprecipitation analysis of SIN3A in A) 293T whole cell lysate of SIN3A protein expressing 293T cells, B) control of pre-immune rabbit IgG, C) immunoprecipitation of SIN3A protein with SIN3A polyclonal antibody (Product # PA5-85554) using A) 30 µg, B)3 µg of sample at a dilution of 1:1000. Sample was then incubated with HRP-conjugated anti-rabbit IgG secondary antibody. Prior to incubation with primary antibody, the sample was separated on 5% SDS-PAGE.
