Western blot analysis of Carbonic Anhydrase IX Recombinant Protein using Product # PA5-19441, Carbonic Anhydrase IX primary antibody at a dilution of 1 µg/mL. Blot treated with a secondary HRP-conjugated Goat polyclonal anti-Rabbit antibody was used at a dilution of 1:3000.

Western blot was performed using Anti-Carbonic Anhydrase IX Polyclonal Antibody (Product # PA5-19441) and ~ 50-55 kDa bands corresponding to CA9 was observed across cell lines tested and increased upon Deferoxamine and Cobalt Chloride treatment in HeLa along with an uncharacterized band (*) at ~35 kDa. Whole cell extracts (30 µg lysate) of U-87 MG (Lane 1), HeLa (Lane 2), HeLa treated with Deferoxamine (100uM for 24 Hours) (Lane 3) and HeLa treated with Cobalt Chloride (200uM for 24 Hours) (Lane 4) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:250 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
