Experimental details: Western blot analysis of HSP90 alpha/beta: purified HSP alpha (lane 1), purified HSP beta (lane 2), MCF-7 lysate (lane 3) and MCF-7 treated with UV (lane 4). Samples were probed with anti-HSP90 alpha/beta monoclonal antibody (Product # MA1-10372).

Experimental details: Knockdown of HSP90 was achieved by transfecting U-87 MG cells with HSP90 specific siRNAs (Silencer® select Product # s6994, s6995). Western blot analysis (Fig. a) was performed using whole cell extracts from the HSP90 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with HSP90 Polyclonal Antibody (Product # MA1-10372, 1 µg/mL dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to HSP90.

Experimental details: Western blot analysis was performed on whole cell extracts (30 µg lysate) of A-431 (Lane 1), U-87 MG (Lane 2), HEK 293T (Lane 3), HeLa (Lane 4), K562 (Lane 5) and NIH/3T3 (Lane 6). The blot was probed with Anti-HSP90 Polyclonal Antibody (Product # MA1-10372, 1 µg/mL dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/mL, 1:4000 dilution). A 80 kDa band corresponding to HSP90 was observed across the cell lines tested.
