- Western Blot Analysis: Untreated and Insulin-treated (30 min) (Lanes 1 and 2 respectively) 293 cell lysates were resolved by electrophoresis, transferred to PVDF and probed with anti-phospho-IRS1 (Ser616) (1/1,000). Peptide blocking was performed using insulin-treated 293 cell lysates. Lanes 3 was unblocked. Lanes 4 and 5 were peptide blocked with phosphorylated or non-phosphorylated IRS1 (Ser616) peptides, respectively.Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates phosphorylated IRS1 (Ser616).
