- Knockdown of Reticulon-4 was achieved by transfecting U-87 MG with Reticulon-4 specific siRNAs (Silencer® select Product # s32768, s32766). Western Blot analysis (Fig. a) was performed using Membrane enriched extracts from the Reticulon-4 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2), and untransfected cells (lane 1). The Blot was probed with Nogo-B Recombinant Rabbit Monoclonal Antibody (JM02-34) (Product # MA5-32763, 1:1,000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution). Densitometric analysis of this western Blot is shown in histogram (Fig. b). The decrease in signal upon siRNA mediated knockdown confirms that the antibody is specific to Reticulon-4.

- Western Blot was performed using Anti-Nogo-B Recombinant Rabbit Monoclonal Antibody (JM02-34) (Product # MA5-32763) and 40-50 kDa bands corresponding to Reticulon-4 was observed across all the cell lines and tissue lysates tested. An ~200 kDa band was also observed in HUVEC cell line that corresponds to Nogo- A. Membrane enriched extracts (30 µg lysate) of HUVEC (Lane 1), HAEC (Lane 2), BJ (Lane 3), Hep G2 (Lane 4), Neuro-2a (Lane 5), C2C12 (Lane 6), THP-1 (Lane 7) as well as Mouse Prostate (Lane 8) and Mouse Liver tissue lysates (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
