- Description
- Polymeric or monomeric Ubiquitin exists free (unanchored) or covalently attached to its ligand. Ubiquitin is conjugated to its target via an isopeptide bond either as a monomer or as one of two polymeric forms. Ubiquitin monomers in one type of polymer are covalently linked via various Lys residues (polyubiquitin). The other type of ubiquitin polymer involves monomers that are linked linearly via an initiator Met (linear polyubiquitin). Polyubiquitin’s functions depend on which Lys residue is involved in cross-linking the protein. K63-linked polyubiquitin chains exhibit nonproteolytic function in vivo which includes kinase/phosphatase activation, protein trafficking, endocytosis, DNA-damage responses, as well as in signaling processes which lead to the activation of the transcription factor NF-κ-B.

- Western Blot showing specificity of clone HWA4C4. Lane 1 & 5: Molecular weight marker; Lanes 2 & 6: HeLa lysate (negative control); Lanes 3 & 7: Poly-Ub(3-7)-K63; Lanes 4 & 8: Poly-Ub(1-7)-K48. Blots were incubated with either clone HWA4C4 or Mouse IgG2a isotype control at 1 µg/ml.
