- CHO-7 cells were cultured in the absence (I) or presence (S) of cholesterol. Equivalent aliquots of chromatin from each sample containing ~50 µg of total protein were subjected to analysis by SDSPAGE and immunoblot. After transfer, the membrane was probed using anti-NF-YA as the primary antibody at a 1/1000 dilution, followed by reaction with HRP conjugated Goat-anti-Rabbit IgG [H&L]. Signal was developed using an ECL kit followed by autoradiography using a 30'' exposure. The antibody recognizes NF-YA as a doublet of ~ 42 kDa in size.

Secondary antibody: Peroxidase rabbit secondary antibody at 1/10,000 for 45 min at RT. Localization: NF-Y is nuclear. Staining: NF-Y subunit A as precipitated red signal with hematoxylin purple nuclear counterstain.
