- Knockdown of DVL2 was achieved by transfecting U-87 MG with DVL2 specific siRNAs (Silencer® select Product # s4398, s4397). Western blot analysis (Fig. a) was performed using whole cell extracts from the DVL2 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with DVL2 Monoclonal Antibody (OTI7C7) (Product # MA5-26816, 1:1000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to DVL2.

- Western blot was performed using Anti-DVL2 Monoclonal Antibody (OTI7C7) (Product # MA5-26816) and a band at 90 kDa corresponding to DVL2 was observed across the cell lines tested. Whole cell extracts (30 µg lysate) of HCT 116 (Lane 1), Raji (Lane 2), U-87 MG (Lane 3), Jeko-1 (Lane 4), NIH/3T3 (Lane 5) and MDA-MB-231 (Lane 6) were electrophoresed using Novex® NuPAGE® 4-12% % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunohistochemistry was performed on paraffin-embedded adenocarcinoma of human endometrium tissue. To expose target proteins, 1 mM EDTA in 10mM Tris, pH8.5, 120°C for 3min was used. Following antigen retrieval, tissues were probed with a DVL2 monoclonal antibody (Product # MA5-26816).
