- Western Blot using SNRPB Polyclonal Antibody (Product # PA5-27559). Whole cell extract (30 µg) was separated by 12% SDS-PAGE, and the membrane was blotted with SNRPB Polyclonal Antibody (Product # PA5-27559) diluted at 1:1,000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

- Knockdown of SNRPB was achieved by transfecting A-431 cells with SNRPB specific siRNAs (Silencer® select Product # s13221, s224659). Western blot analysis (Fig. a) was performed using modified whole cell extracts (1% SDS) from the SNRPB knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with SNRPB Polyclonal Antibody (Product # PA5-27559, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to SNRPB.

- Western blot analysis was performed on modified whole cell extracts (1% SDS) (30 µg lysate) of A-431 (Lane 1), NTERA-2 (Lane 2), Hep G2 (Lane 3), MOLT-4 (Lane 4), HEK-293 (Lane 5), K-562 (Lane 6), T-47D (Lane 7), SH-SY5Y (Lane 8) and HeLa (Lane 9). The blot was probed with SNRPB Polyclonal Antibody (Product # PA5-27559, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 25kDa band corresponding to SNRPB was observed across the cell lines tested.

- Immunofluorescent analysis of SNRPB in paraformaldehyde-fixed HeLa cells using a SNRPB polyclonal antibody (Product # PA5-27559) (Green) at a 1:500 dilution. Alpha-tubulin filaments were labeled with Product # PA5-29281 (Red) at a 1:2000.

- Detection of binding of endogenous SNRPB protein to specific RNA using Anti-SNRPB Antibody: RNA Immunoprecipitation (RIP) was performed using Anti-SNRPB Rabbit Polyclonal Antibody (Product # PA5-27559, 5 µg) on clarified whole cell lysate from 4 million Hep G2 cells. Normal Rabbit IgG was used as a negative IP control. Immunoprecipitated RNA was purified by RiboPure™ RNA Purification Kit (Product # AM1924) and analyzed by RT-PCR using the Power SYBR® Green RNA-to-CT™ 1-Step Kit (Product # 4389986) with primer pairs over U1 snRNA (positive) and 18S rRNA, GAPDH mRNA (negative). Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method.
