Western blot was performed using Anti-PLA2G4A Polyclonal Antibody (Product # PA5-19324) and a ~85 kDa band was observed across the panel tested except IMR-32 and Reh. Whole cell extracts (30 µg lysate) of THP-1 (Lane 1), HeLa (Lane 2), A549 (Lane 3), NIH/3T3 (Lane 4), IMR-32 (Lane 5) and Reh (Lane 6) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Rabbit anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Flow cytometric analysis of PLA2G4A in A549 cells using a polyclonal antibody (Product #PA5-19324). A549 cells (blue line) were paraformaldehyde fixed and permeabilized with 0.5% Triton. The primary antibody was incubated for one hour (10 µg/mL) followed by an Alexa Fluor 488 secondary antibody (1 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by an Alexa Fluor 488 secondary antibody.
