- Western blot analysis of Lactate Dehydrogenase A (LDHA) was performed by loading 30 µg of normal or diabetic human melanocyte whole cell lysates, treated with 0.4mM palmitate for 0, 4 or 24 hours, per well onto an SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk for 1 hour. The membrane was probed with an LDHA polyclonal antibody (Product # PA5-23036) at a dilution of 1:1000 at 4C overnight, washed in PBST, and probed with an HRP-conjugated anti-rabbit IgG secondary antibody at a dilution of 1:10,000 for 1 hour at room temperature. - Western blot analysis was performed on whole cell extracts (30 µg lysate) of U-87 MG (Lane 1), PC-3 (Lane 2), A-431 (Lane 3), MCF-7 (Lane 4), DU 145 (Lane 5) MDA-MB-231 (Lane 6) and PANC-1 (Lane 7). The blot was probed with Anti-LDHA Polyclonal Antibody (Product # PA5-23036, 1 µg/mL dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 35 kDa band corresponding to LDHA was observed across the cell lines tested.
