Description

Ligufalimab (also known as AK117) is a humanized IgG4 monoclonal antibody that specifically targets CD47, a cell-surface protein known as a ¡°don¡¯t eat me¡± signal. Its IgG4 isotype is selected to minimize effector functions like antibody-dependent cellular cytotoxicity (ADCC). A key distinguishing feature of ligufalimab is its engineered binding property that does not induce red blood cell (RBC) agglutination, a common side effect associated with anti-CD47 antibodies. By blocking the interaction between CD47 and its macrophage receptor SIRP¦Á, it selectively promotes the phagocytosis of tumor cells by macrophages. It is under clinical investigation for the treatment of advanced solid tumors and hematologic malignancies.
