- MRP1 is a 190 kD integral membrane phosphoglycoprotein. It is a member of the human ATP-binding cassette transporter multidrug resistance protein1 (MRP1), encoded by ABCC1. MRP1 is expressed in most normal tissues, with higher levels in the testes, lung, skin, small intestine, colon, kidney, placenta, peripheral blood mononuclear cells, and skeletal muscle. ABCC1 encodes plasma membrane proteins that have important physiological transport functions and mediate a variety of xenobiotics against their concentration gradients. MRP1 plays an important role in the extrusion of drugs from the cell and their over expression can be a cause of failure of anticancer and antimicrobial chemotherapy.

- Human paraffin-embedded colon tissue slices were prepared with a standard protocol of deparaffination and rehydration. Antigen retrieval was done with BioLegend’s Tris-Buffered Saline 20X (1.0M, pH7.4) at 95°C for 40 minutes. This tissue was washed with PBS/0.05% Tween 20 twice for five minutes, permeabilized with 0.5% Triton X-100 for ten minutes, and blocked with 5% FBS and 0.2% Gelatin for 30 minutes. Then, the tissue was stained with 5 µg/mL of anti-human ABCC1 (clone QCRL-3) Alexa Fluor® 594 (red) and Pan-Cytokeratin Alexa Fluor ® (green) at 4°C overnight. Nuclei were counterstained with DAPI (blue). This image was captured with a 10X objective.
