Western blot analysis of ALY in HeLa cell extract using an ALY monoclonal antibody (Product # MA1-26754) at a dilution of 0.4 µg/mL followed by a goat anti-mouse peroxidase conjugate (lane 1, 2) and a chemiluminescent substrate.

Experimental details

Western blot analysis of ALY was performed by loading 5 µL of HeLa and HEK293T cell lysates obtained by lysing cells grown to confluency in a 15cm dish in 500 µL of lysis buffer per well onto an SDS-PAGE gel. Proteins were transferred to a nitrocellulose membrane and blocked overnight with 5% milk in PBST. The membrane was probed with an ALY monoclonal antibody (Product # MA1-26754) at a dilution of 1:2500 for 1 hour at room temperature, washed in PBST, and probed with an HRP-conjugated anti-mouse IgG secondary antibody at a dilution of 1:5000 for 1 hour at room temperature. Experimental details

Immunofluorescent analysis of ALY (red) in HeLa cells. Cells fixed in 4% paraformaldehyde were permeabilized with 0.1% Triton X-100 for 5-10 minutes at room temperature and washed 3 times with PBS containing 1% Fetal Calf Serum (wash buffer) at room temperature. Cells were probed with an ALY monoclonal antibody (Product # MA1-26754) at a dilution of 1:500 for 1 hour at room temperature, washed with wash buffer, and incubated with a fluorescently-conjugated anti-mouse IgG secondary antibody at a dilution of 1:200 for 1 hour at room temperature. Nuclei (blue) were stained with DAPI. Images were taken on an LSM780 (Zeiss Microscope) at 63x magnification. Data courtesy of the Innovators Program.
