- MA1-870 detects RPA p34 in human and rat cells. MA1-870 has been successfully used in Western blotting, immunofluorescence and immunohistochemistry (paraffin) procedures. By Western blot it detects a 34 kDa protein representing RPA p34. The MA1-870 immunogen is the RPA p34 subunit isolated from HeLa cells.

- Knockdown of RPA2 was achieved by transfecting Caco-2 with RPA2 specific siRNAs (Silencer® select Product # s12130, s12132). Western blot analysis (Fig. a) was performed using whole cell extracts from the RPA2 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with RPA2 Monoclonal Antibody (MA34) (Product # MA1-870, 1:500 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/ml 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to RPA2.

- Western blot analysis was performed on modified whole cell extracts (1% SDS) (30 µg lysate) of HeLa (Lane 1), MDA-MB-231 (Lane 2), Hep G2 (Lane 3), MCF7 (Lane 4), Jurkat (Lane 5), HEK 293 (Lane 6) and COS7 (Lane 7). The blot was probed with Anti-RPA2 Monoclonal Antibody (Product # MA1-870, 1:500 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/ml, 1:4000 dilution). A 32 kDa band corresponding to RPA2 was observed across the cell lines tested.

Cells were probed without (control) or with a RPA34 monoclonal antibody (Product # MA1-870) at a dilution of 1:200 overnight at 4 C, washed with PBS and incubated with a DyLight-488 conjugated secondary antibody (Product # 35503). To expose target proteins, heat induced antigen retrieval was performed using 10mM sodium citrate (pH6.0) buffer, microwaved for 8-15 minutes. Following antigen retrieval tissues were blocked in 3% BSA-PBS for 30 minutes at room temperature and probed with a RPA34 monoclonal antibody (Product # MA1-870) at a dilution of 1:20 or without primary antibody (negative control) overnight at 4°C in a humidified chamber. Tissues were washed with PBST and endogenous peroxidase activity was quenched with a peroxidase suppressor. Detection was performed using a biotin-conjugated secondary antibody and SA-HRP, followed by colorimetric detection using DAB. Tissues were counterstained with hematoxylin and prepped for mounting.
