- Figure 3 The immunotoxin hD7-1(VL-VH)-PE40 induces apoptosis in PSMA-positive cells by reducing the expression of Mcl-1 and Bcl2A1. PC cells were incubated for different time points with 1 nM immunotoxin. After cell lysis, expression of the anti-apoptotic Bcl-2 proteins Bcl-2, Bcl-xl, Mcl-1, Bcl-w, and Bcl2A1, the pro-apoptotic Bcl-2 proteins Bax and Bak, as well as the activation of caspase-3, and -8, and the cleavage of PARP were determined by Western blot. ss-actin was detected as loading control.
