Western blot was performed using Anti-ERG Polyclonal Antibody (Product # PA5-49681) and a ~52kDa band corresponding to ERG was observed across cell lines tested. Other isoforms corresponding to ~42kDa and ~ 75kDa were observed in MCF-7 and MOLT-4 respectively. Modified whole cell extracts (1% SDS) (30 µg lysate) of HeLa (Lane 1), Jurkat (Lane 2), MOLT-4 (Lane 3) and MCF-7 (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Western blot was performed using Anti-ERG Polyclonal Antibody (Product # PA5-49681) and a ~42 kDa band corresponding to ERG was observed across all the tissue extracts tested except Mouse Spleen which is reported to be negative. Tissue extracts (30 µg lysate) of Mouse Lung (Lane 1), Mouse Liver (Lane 2), Mouse Brain (Lane 3), Mouse Kidney (Lane 4) and Mouse Spleen (Lane 5) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752).
