- Carboxyl-terminal PDZ ligand of neuronal nitric oxide synthase(nNOS) protein or CAPON (UniProt O75052), also known as Nitric oxide synthase (NOS) 1 adaptor protein, C-terminal PDZ ligand of nNOS, C-terminal PDZ domain ligand of nNOS, Ligand of nNOS with carboxyl-terminal PDZ domain) is a cytosolic protein encoded by the NOS1AP gene (Entrez Gene ID 9722) in human. NOS1AP protein (CAPON) contains a C-terminal PDZ-binding domain and an N-terminal phosphotyrosine binding (PTB) domain that mediates its interactions with nNOS and the small monomeric G protein Dexras1, respectively. Originally it was characterized as a competitor against PSD95 for nNOS interaction, but later evidence indicates that nNOS-CAPON association contributes to the modulation of anxiety-related behavior via regulating Dexras1-ERK signaling. CAPON is also reported to co-localize with the L-type calcium channel and the inwardly rectifying potassium channel Kir3.1 in murine cardiac myocytes. In addition, CAPON is significantly upregulated in mice with dystrophic cardiomyopathy, implicating a role of NOS1AP protein on cardiac arrhythmias, most likely through S-nitrosylation of L-type calcium channel and potassium channels.
