- Western blot was performed using Anti-DEK Polyclonal Antibody (Product # PA5-63488) and a 50 kDa band corresponding to DEK was observed in all the tested cell lines and the expression was lower in MCF 10A when compared to MDA-MB-231. Modified whole cell lysates (1% SDS) (30 µg lysate) of HeLa (Lane 1), PANC-1 (Lane 2), IMR-32 (Lane 3), PC-3 (Lane 4), Jurkat (Lane 5), MCF 10A (Lane 6) and MDA-MB-231 (Lane 7) were electrophoresed using NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blots were probed with the primary antibody (0.4 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
