Experimental details

Western blot analysis of CTPS in different lysates. Exposure time: 1 minute; 10% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. Samples were incubated in CTPS Monoclonal antibody (Product # MA5-44804) using a dilution of 1:500 in 5% NFDM/TBST at room temperature for 2 hours followed by Goat Anti-Rabbit IgG - HRP secondary antibody at a dilution of 1:200,000 for 1 hour at room temperature. Lane 1: NIH/3T3 cell lysate (10 µg/Lane); Lane 2: Mouse spleen tissue lysate (20 µg/Lane). Predicted band size: 67 kDa. Observed band size: 75 kDa.

Experimental details

Flow cytometry of CTPS in THP-1 cells. Cells were fixed and permeabilized then stained with CTPS Monoclonal antibody (Product # MA5-44804) using a dilution of 1 µg/mL (red) at 4℃ for an hour followed by iFluor™ 488 conjugate-Goat anti-Rabbit IgG secondary antibody at a dilution of 1:1,000 for 30 minutes at 4℃. Rabbit IgG Isotype Control (green). Unlabeled sample was used as a control (cells without incubation with primary antibody; black).
