- Western blot analysis of MOG in Lane 1: human HEK293 whole cell lysate, Lane 2: human HK-2 whole cell lysate, Lane 3: human SGC-7901 whole cell lysate, Lane 4: rat brain tissue lysate, Lane 5: mouse brain tissue lysate. Electrophoresis was performed with 5-20% SDS-PAGE gel (70V, Stacking gel; 90V Resolving gel, Time: 2-3 hours), transferred to a nitrocellulose membrane and blocked using 5% Non-fat Milk/TBS (1.5 hrs at room temperature). Samples were incubated with MOG polyclonal antibody (Product # PA5-95602) using a 0.5 µg/mL dilution, followed by a goat anti-rabbit IgG-HRP at a dilution of 1:10,000, and developed with enhanced chemiluminescence (ECL).

- Western blot analysis of Myelin oligodendrocyte glycoprotein in, Lane 1: rat brain tissue lysates, Lane 2: mouse brain tissue lysates, . Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50 µg of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. The membrane was blocked with 5% Non-fat Milk/ TBS for 1. 5 hour at RT. The membrane was incubated with MOG Polyclonal Antibody (Product # PA5-95602) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0. 1% Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1. 5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit. A specific band was detected for Myelin oligodendrocyte glycoprotein at approximately 26KD. The expected band size for Myelin oligodendrocyte glycoprotein is at 28KD.

- Western blot was performed using Anti-MOG Polyclonal Antibody (Product # PA5-95602) and a 27 kDa band corresponding to Myelin-oligodendrocyte glycoprotein was observed Mouse and Rat Brain and Cerebellum but not in Mouse and Rat Heart and Liver. Tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Rat Brain (Lane 2), Mouse Cerebellum (Lane 3), Rat Cerebellum (Lane 4), Mouse Heart (Lane 5), Rat Heart (Lane 6), Mouse Liver (Lane 7) or Rat Liver (Lane 8) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
