- Western blot analysis of ATRX was performed by loading 25μg/mL of lysate without (lane 1) or with treatment of Dox for 72 hours in reducing sample buffer (20 mM Tris-HCl (pH 7.5) 150 mM NaCl, 1 mM Na2EDTA, 1 mM EGTA, 1% NP-40, 1% sodium deoxycholate, 2.5 mM sodium pyrophosphate, 1 mM β-glycerophosphate, 1 mM Na3VO4, 1 µg/mL leupeptin) onto an 8% polyacrylamide gel. Proteins were transferred to PVDF membrane under semi-dry transfer conditions, constant amperage at 45mA per gel for 90 minutes. Targets were detected at approximately ~100 kDa for HIRA, above 250 for ATRX and between 75 and 100 kDa for DAXX using polyclonal antibodies (Product # PA5-51656 for ATRX, Product # PA5-82362 for DAXX and Product # PA5-97065 for HIRA) at a dilution of 1:1000 in TBST containing 5% non-fat dry milk, followed by incubation with a secondary antibody at a dilution of 1:10,000.
