Western blot was performed using Anti-IRAK2 Polyclonal Antibody (Product # PA5-19917) and a 70 kDa band corresponding to Interleukin-1 receptor-associated kinase-like 2 was observed across cell lines tested. Whole cell extracts (40 µg lysate) of U-937 (Lane 1) and U-937 treated with 50 nM of TPA and Ionomycin for 48 hours were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 μg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:5,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescentdetection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Uncharacterized bands (*) were observed at around ~40 and 55 kDa.
