Detection of human and mouse DDX3 by western blot. Samples: Whole cell lysate (50 µg) from HeLa, HEK293T, Jurkat, TCMK-1, and NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-DDX3 antibody (Product # A300-474A; lot A300-474A-2) used for WB at 0.04 µg/mL. Detection: Chemiluminescence with an exposure time of 3 seconds.

Experimental details

Detection of human DDX3 by western blot and immunoprecipitation. Samples: A. Whole cell lysate from HeLa (5, 15 and 50 µg) and Ramos (50 µg) cells. B. Whole cell lysate (1 mg/IP; 1/4 of IP loaded/lane) from HeLa cells. Antibodies: Affinity purified rabbit anti-DDX3 antibody BL1649 (Cat. No. A300-474A) used at 0.04 µg/ml (A) and 0.1 µg/ml (B) for western blot and at 3 µg/mg lysate for IP (B). DDX3 was also immunoprecipitated using rabbit anti-DDX3 antibodies BL1650 (Cat. No. A300-475A) and BL1651 (Cat. No. A300-476A) at 3 µg/mg lysate. Detection: Chemiluminescence with an exposure time of 10 seconds (A and B).

Antibodies: Affinity purified rabbit anti-DDX3 antibody (Product # A300-474A; lot A300-474A-2) used for IP at 6 µg per reaction. DDX3 was also immunoprecipitated by a previous lot of this antibody (lot A300-474A-1) and rabbit anti-DDX3 antibody (Product # A300-476A). For blotting immunoprecipitated DDX3 (Product # A300-474A) was used at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 1 seconds.
