Description

Description: This EP10 monoclonal antibody reacts with human Synaptophysin, which is also known as Major Synaptic Vesicle Protein p38. This 38-kDa integral membrane protein is expressed on neuronal presynaptic vesicles in the brain, spinal cord, retina, neuromuscular junctions, and adrenal medulla. Synaptophysin is also expressed in neuroendocrine tumors and neuroblastomas. Synaptophysin has been reported to play a role in synaptic vesicle trafficking.

Western blot was performed using Anti-Synaptophysin Monoclonal Antibody (EP10), eBioscience™ (Product # 14-6525-82) and a 38 kDa band corresponding to Synaptophysin was observed in SH-SY5Y and IMR-32, but not in SK-BR-3 and BJ. Whole cell extracts (40 µg lysate) of SH-SY5Y (Lane 1), IMR-32 (Lane 2), SK-BR-3 (Lane 3), BJ (Lane 4) were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel (Product # NP0342BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (5 µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

Immunohistochemistry of formalin-fixed paraffin embedded human cerebellum using 5 µg/mL of Mouse IgG1 kappa Isotype Control (Product # 14-4714-82) (left) or Anti-Human Synaptophysin Purified (right) followed by Anti-Mouse IgG Biotin and DAB visualization.Nuclei are counterstained with hematoxylin.
