Antibody: Affinity purified rabbit anti-PCM1 antibody (Product # A301-149A; lot A301-149A-2) used for WB at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 75 seconds.

Experimental details

Detection of human and mouse PCM1 by western blot (h&m) and immunoprecipitation (h). Samples: Whole cell lysate from HeLa (5, 15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 µg) and mouse NIH3T3 (M; 50 µg) cells. Antibodies: Affinity purified rabbit anti-PCM1 antibody A301-149A used for WB at 0.04 µg/ml (A) and 0.1 µg/ml (B) and used for IP at 3 µg/mg lysate. PCM1 was also immunoprecipitated by rabbit anti-PCM1 antibody A301-150A, which recognizes a downstream epitope. Detection: Chemiluminescence with exposure times of 30 seconds (A) and 10 seconds (B).

Detection of human PCM1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-PCM1 antibody (Product # A301-149A; lot A301-149A-2) used for IP at 6 µg per reaction. PCM1 was also immunoprecipitated by a previous lot of this antibody (lot A301-149A-1) and rabbit anti-PCM1 antibody (Product # A301-150A). For blotting immunoprecipitated PCM1, Product # A301-149A was used at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 10 seconds.
