- Western blot analysis of DNA-PKcs was performed by loading 30 µg of MiaPaCa-2 lysate (lane 1), 30 µg of OVCAR-8 lysate (lane 2) and 30 µg of MDA-MB-231 lysate (lane 3) in reducing sample buffer and Page Rµler Protein Ladder (Product # 26616) onto a 6% polyacrylamide gel. Proteins were transferred to PVDF membrane using the wet transfer method. Membrane was blocked in Super Block Blocking Buffer (Product # 37515). DNA-PKcs was detected at approximately 469 kDa using a DNA-PKcs monoclonal antibody (Product # MA5-13404) at a dilution of 1:1000 in 5% BSA, followed by a goat anti-mouse IgG DyLight 800 conjugated secondary antibody (Product # SA5-35521) at a dilution of 1:7500. - Western blot analysis of DNA-PKcs was performed by loading ~300,000 cell equivalents per lane in 1X LDS reducing sample buffer (Product # NP0007) and molecular weight ladder onto a 3-8% Tris-Acetate protein gel (Product # EA0375BOX). Proteins were transferred to a PVDF membrane at 100 mA overnight. Membrane was blocked in 5% milk/TBST for 1 hour. DNA-PKcs was detected using a monoclonal antibody (Product # MA5-13404) diluted 1:1000 at 4°C, overnight. Membrane was then probed with anti-mouse secondary antibody at a dilution of 1:5000. - Immunofluorescence analysis of DNA-PKcs was performed using 70% confluent log phase A549 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with DNA-PKcs (18-2 + 25-4 + 42-psc) Mouse Monoclonal Antibody (Product # MA5-13404) at 2µg/ml in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjµgate (Product # A28175)at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. - Flow cytometry analysis of DNA-PKcs was done on HeLa cells. Cells were fixed with 70% ethanol for 10 minutes, permeabilized with 0.25% Triton™ X-100 for 20 minutes, and blocked with 5% BSA for 30 minutes at room temperature. Cells were labeled with DNA-PKcs Mouse Monoclonal Antibody (Product # MA5-13404, red histogram) or with mouse isotype control (pink histogram) at 3-5 µg/million cells in 2.5% BSA. After incubation at room temperature for 2 hours, the cells were labeled with Alexa Fluor® 488 Rabbit Anti-Mouse Secondary Antibody (Product # A11059) at a dilution of 1:400 for 30 minutes at room temperature. The representative 10,000 cells were acquired and analyzed for each sample using an Attune® Acoustic Focusing Cytometer. The purple histogram represents unstained control cells and the green histogram represents no-primary-antibody control..
