- Western blot analysis of DNA-PKcs was performed by loading 30 µg of MiaPaCa-2 lysate (lane 1), 30 µg of OVCAR-8 lysate (lane 2) and 30 µg of MDA-MB-231 lysate (lane 3) in reducing sample buffer and Page Rµler Protein Ladder (Product # 26616) onto a 6% polyacrylamide gel. Proteins were transferred to PVDF membrane using the wet transfer method. Membrane was blocked in Super Block Blocking Buffer (Product # 37515). DNA-PKcs was detected at approximately 469 kDa using a DNA-PKcs monoclonal antibody (Product # MA5-13244) at a dilution of 1:1000 in 5% BSA, followed by a goat anti-mouse IgG DyLight 800 conjugated secondary antibody (Product # SA5-35521) at a dilution of 1:7500.
