- Figure 7 The Tid1-galectin-7-MMP-9 pathway might contribute to cell migration and invasion in NUGC-3 gastric cancer cells. ( A ) The effect of Tid1 knockdown on galectin-7 expression was determined by Western blot. ( B ) The effect of Tid1 knockdown on galectin-7 protein stability was determined by cycloheximide chase analysis. After knockdown of Tid1 by siRNA, the gastric cancer cells were reseeded and treated with cycloheximide for the indicated time periods. ( C ) Kaplan-Meier survival analyses were performed using the Kaplan-Meier plotter online database ( http://kmplot.com/analysis/ ) []. The results showed the effect of galectin-7 gene expression on overall survival (OS, left) and progression-free survival (PFS, right) in gastric cancer patients. ( D , E ) The effects of Tid1 knockdown on gene expression and activity of MMP-9 were determined by ( D ) RT-real-time PCR and ( E ) gelatin zymography analysis. The efficiency of Tid1 knockdown was confirmed by Western blot and RT-real-time PCR.
