Experimental details: Western Blot analysis of Flotillin 1 was performed by separating 30 µg of various whole cell extracts by 10% SDS-PAGE. Proteins were transferred to a membrane and probed with a Flotillin 1 Polyclonal Antibody (Product # PA5-28118) at a dilution of 1:1000 and a HRP-conjugated anti-rabbit IgG secondary antibody.

Experimental details: Western blot was performed using Anti- Flotillin 1 Polyclonal Antibody (Product # PA5-28118) and a 47kDa band corresponding to Flotillin 1 was observed across all the cell lines and tissue lysates tested except THP1 which is reported negative for Flotillin 1 expression. An uncharacterized band was observed at ~35KDa in A549 sample. Whole cell extracts (30 µg lysate) of HEK-293 (Lane 1), MCF7 (Lane 2), MDA-MB-231 (Lane 3), HeLa (Lane 4), A549 (Lane 5), THP-1 (Lane 6), tissue extracts of Mouse Lung (Lane 7), Mouse Brain (Lane 8) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blots were probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence Goat Anti-Rabbit IgG Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005)..
